A systems approach to Parkinson’s disease

چکیده
We describe a systems approach to Parkinson’s disease in which an integrative mathematical model of the brain energy metabolism is used as the basis for in–silico investigation of possible disease mechanisms. The mathematical model allows both the analysis of dynamical and structural implications of biological factors implicated in disease causation, and the potential role of a compromised brain energy metabolism as factor that enables pathologies associated with the disease to develop. Using a recently developed mathematical model of brain energy metabolism, we use simulations of brain energy behaviour to illustrate how a weakened energy metabolism might create a cellular environment where pathogenic mechanisms that are normally held in check by an efficient energy metabolism could potentially flourish. Simulations of the brain energy metabolism are presented which illustrate how both very long term deterioration of metabolism, and cumulative stress from energy transients, may create a predisposition to Parkinson’s disease. The focus of this study is Parkinson’s disease, but we note that our energy metabolism modelling and analysis framework may also form a basis for a systems approach to other age–related neurodegenerative conditions. ∗The Hamilton Institute National University of Ireland Maynooth, County Kildare, Ireland.

The Effects of Bilateral Subthalamic Nucleus Stimulation on Cognitive and Neuropsychiatric Functions in Parkinson’s Disease: A Case-Control Study
Introduction: Parkinson’s disease is one of the most disabling diseases which by electrode implantation and stimulation of subthalamic nucleus (STN), much progress has been made in the treatment of drug resistant patient. This new method of neurosurgery may have some neuropsychological side effects on the patients. The main aim of this study is to evaluate the effects of this kind of treatment ...

Depression is common and may occur at any stage in the long history of Parkinson’s disease, markedly affecting a patient’s threshold to discomfort, but it is particularly difficult to assess before clear clinical features are evident. The rate of depression in patients with Parkinson’s disease vary, partly because of differences in the definition of depression, difficulties in distinguishing be...

متن کاملDiagnosis of Parkinson’s Disease in Human Using Voice Signals
A full investigation into the features extracted from voice signals of people with and without Parkinson’s disease was performed. A total of 31 people with and without the disease participated in the data collection phase. Their voice signals were recorded and processed. The relevant features were then extracted. A variety of feature selection methods have been utilized resulting in a good perf...
متن کاملMedical Food to Stop the Progression of Parkinson’s Disease
No progress has been made in the development of drugs to stop the progression of Parkinson’s Disease. Here the author has presented a novel approach to stopping the disease using a dietary supplement primarily composed of Mannitol. In vivo animal studies have shown that Mannitol was able to break up alpha-synuclein clusters and restore functioning in transgenic drosophila and mice. The author, ...
